CRISPR-Cas9 ("Mr. Sandman" Parody) | A Capella Science

eric3579 says...

CRISPR-Cas9
Bring me a gene
Encoding for a specific protein
Make a few snips at this coded locus
You work so well inside a streptococcus
Cas9
I'm so alone
Without your scissors in my chromosome
Cut me up and do it clean
CRISPR-Cas9 bring me a gene

CRISPR-Cas9
Keep me a gene
A viral sequence you've already seen
Chopped into bits and stored as genomic
With clustered repeats
That are palindromic
Cas9
Bind with this code
Use it to target infections of old
Immunized like a vaccine
CRISPR-Cas9 keep me a gene

CRISPR-Cas9
Cut me a gene
With a precision that I've never seen
Unzip a strand and interrogate it
Seek out your sequence until you locate it
Cas9
Lock into place
And do your job as endonuclease
Chop just like a guillotine
CRISPR-Cas9 cut me a gene

Snip snap!
CRISPR-Cas9
CRISPR-Cas9

CRISPR-Cas9
Bring me a gene
By commandeering my repair routine
A strand to match your severed location
For some homologous recombination
Cas9
Cheap and precise
Rewriting genomes from microbes to mice
And soon the humble human being
CRISPR-Cas9 bring me a gene

CRISPR-Cas9
Give us a gene
Give us a miracle like that one Nazarene
‘Cause giving the lame their legs and the blind their sight is
In view for dystrophy and retinitis
But CRISPR-Cas9
What if you fall
Outside our power and inside us all
That really could incite a scene

When this terrible wonderful power unsettling
Opens the door to unethically meddle
Is ev’ry congenital malady bettered
Sufficient to warrant genetics unfettered
To modify man in the manner of Gattaca
Raise up a mammoth or make a rattata
Dramatical medical means to eradicate aging
Or cancer or make a fanatic
A mass epidemic a weapon nefarious
Single mosquito to wipe out malaria
Send in a viral infection to ferry a
Cure to the cells of an HIV carrier
Freed of disease as we're free to uncover
What nature and accident failed to discover
And free to be other than
All that we ever have been

CRISPR-Cas9
CRISPR-Cas9

Oh CRISPR-Cas9
Bring us a gene
You wondrous ribonucleoprotein
You have the power to vanquish or save us
Who would have thought that the microbe that gave us
Cas9
S. pyogenes
The source of strep and flesh-eating disease
Housed this marvellous machine
Full of uses great and obscene
CRISPR-Cas9 bring us
Please don't sting us
Cas9 bring us a gene

With adenine
And thiamine
Incite a scene
Cas9 bring us a gene!

The Irish Island Taking In Anti-Trump Refugees

Can You Solve the Bridge Riddle?

dannym3141 says...

By a process of elimination - you know you can't do it with just the 1 minute guy ferrying everyone because 10+5+2=17 so no time for 2 return trips even at 1 minute. That made me realise two big numbers had to cross together without a big number ferrying it back. Then i just fiddled until i found a way to bring the lantern back from their trip without using up 5 minutes.

However i spent 15 minutes thinking about it, so me and 2 minutes sprinted across to safety.

It took so long to set up that I didn't try to work it out and simply waited 3 seconds for the answer, which by the way, was very clever!

Anyone try to do the math first?

Going to the fair in China!!

LOL! That would most certainly kill you; some of them are simply outright impossible (the "first ones"). That Ferris Wheel, oh god.

The Onion Looks Back At 'Back To The Future'

poolcleaner says...

Back to the Future.

Married with Children.

The Simpsons.

Ferris Buehler's anarchist ideology towards his and Cameron's upper class parents. Handing over the car keys to the lower class joy riders.

The Goonies going on adventures to prevent a golf course from overtaking their upper class lifestyle.

Was Mikey about to become middle class?! Holy shit. The Goonies is about retaining your social position in order to fund misadventure. And the rise of the middle class Hispanic family! i.e. Rosalita discovering the jewels in the marble bag.

Everything from the '80s involving a family was about -- Anyone? Anyone? -- Voodoo Economics.

That's actually a pretty solid take on it for the Onion. As usual, it's funny because it's true. *promote

Last Week Tonight with John Oliver: Mental Health

yellowc says...

There's not much to it I'm afraid.

Plenty of successful treatments are available. It's simply a matter of funding.

From my experiences in Australia, both close and friends who work in mental health. They're overworked, the pay sucks, there's no budget to hire more staff and the rooms are crowded.

There has been multiple instances when a patient who needs to be admitted, will be left in a bed in emergency and ferried about as needed, while a bed in the mental health ward frees up.

This is the sort of funding mental health workers are dealing with.

If your patient is already at home and their low risk, you'll be told to just try to keep them at home and manageable for as long as possible. You're on waiting list, so just hope nothing happens in the mean time during their relapse.

What people fail to understand, is if all the workers are so stressed out to the point of trying to shuffle just getting some one a place to sleep. How in the hell do they have time to do any sort of actual therapy?

They get criticised for just pushing drugs and pumping people out after 2-3wks and yet no one appreciates that they must. Because they just got a new patient that is severely ill and a person with mild schizophrenia is going to be sent along on their merry way, regardless if they needed another month or two to fully stabilise. They're doing ok and they can just revolve back around when the meds wear off, perhaps they'll have another room then.

It's sad but it's reality and it's not their fault. You can point fingers and demand better this and that and this. Or you can realise just the mere fact that these people have these terribly shitty jobs means they care a whole lot more fucks than you or I ever have or will.

They need more money, simple.

Another Truck Hits That Massachusetts Bridge

lucky760 says...

A big truck like that was in front of me waiting to get on a ferry a few months ago, but they realized there's no way they could get onto it (without sinking it), so they started backing up, and as I started laying on my horn they smashed into my front end.

Big trucks seem to elicit blissful unawareness and overconfidence.

MACKLEMORE & RYAN LEWIS - DOWNTOWN (OFFICIAL MUSIC VIDEO)

moonsammy says...

I don't quite know what to make of it. It's like Uptown Funk with a more 70s / 80s vibe, plus a lanky skeevy drag king (?) who seems to sound like 90s Michael Jackson... sort of ends all Ferris Bueller. Strange.

Blues Brothers - opening scene

poolcleaner says...

Them just out of the '70s, 1980s sensibilities. Jump the river with your cop motor and FIX that cigarette lighter. I need to watch Stripes now so I can watch the sexy military police have sex with the military fuck ups. Then I'll fast forward the decade and watch Ferris Buehler not go to school.

Film in the 80s taught me about anarchy and it taught me well. Oh wait, film as media is inherently anarchistic. Aw, fuck it. Society must just stink.

Clumsy ferry goer almost cut in half

I should mention as a footnote that with the arrival of my little boy, when the missus went back to work I've had to buy a car to help with ferrying him to daycare, etc. Couldn't really put him in the backpack on the bike.

The bike's since been mothballed in the garage. The missus has said that I can ride it again, but only if I show her a current life insurance policy with a minimum value enough to pay off the mortgage.

Guy Hides In Rear Bumper Trying To Cross The Border

Starboard tack does NOT have right of way over a ferry...

robbersdog49 says...

I was wondering about the spinnaker pole too, seemed a bit of a brave move in those conditions! As did completely un-reefed sails. Looks like they'd read the book as far as 'let the sails out to slow down or stop' but not got any further.

One thing they could have done to avoid the collision is just pull the main in, hard. Would have screwed the boat round into the wind and either stopped it there or more likely forced a tack, leaving the boat sailing happily away from the ferry. The guys sailing it just panicked and did nothing. Fortunately it looked like there wasn't too much damage done but this sort of thing bothers me. Ignorance like this kills.

Looks like a Sydney ferry, so *downunder.

That boat was not under control at all. The jib is starting to rip and god only knows why they had the spinnaker pole attached.

SFOGuy says...

Sorry, one more detail; the newspaper story. Ferry altered course to port (left) to try and dodge but there was a reef/rocks and she couldn't move much more---
Had to stop and assist the rather clueless sailors.
